The Museo de las Culturas de Oaxaca occupies the former Santo Domingo de Guzmán convent in Oaxaca City, a colonial building with restored courtyards, chapels, and rooms. Fourteen permanent halls and nine thematic rooms bring together archaeological, historical, and ethnographic collections from across Oaxaca.

The exhibitions begin with hunter-gatherer communities from around 10,000 years ago, including projectile points from the Oaxaca Valley. They continue through the Zapotec rise and Monte Albán, the Mixtec Postclassic period, Spanish conquest, colonial change, independence, the Porfiriato, revolution, and modern Oaxaca. Fifteen ethno-linguistic groups appear through their languages, communal traditions, civic responsibilities, crafts, and traditional knowledge. A central highlight is the treasure from Tomb Seven at Monte Albán, discovered by Alfonso Caso in 1932.

Nine rooms focus on ceramics, goldsmithing, wood carving, leatherwork, music, visual arts, gastronomy, and beverage production. Objects linked to the Dominican friars and archaeological discoveries reveal the history of the Santo Domingo complex itself. Four temporary exhibition halls occupy former spaces such as the refectory, kitchen, and chapels.
